Abstract

Attacus atlas is one of potent silk-producing insect and possesses a significant economic value. A. atlas is part of genus Attacus which inhabits in several quite large areas. A.atlas can be found in Sumatera, Java, Borneo, Halmahera Islands, and Papua. Mostly, species which inhabits in wide spread area, has a high level of diversity. Applied studies on Antheraea dan Samia cynthia ricini showed that geographical differences led to genetic variations occurred upon those species. The aim of this research was to study and analyze the genetic variation A. Atlas, which is obtained from multiple sampling locations, ie Berbah, Dlingo, Ngawen, and Pracimantoro, based on ISSR marker. Analysis of genetic variation performed with 4 ISSR primer, ISSR1, ISSR2, ISSR6, and ISSR7 gave forth high genetic variations among individuals of A.atlas by indicating the average polymorphic percentage of all primer of 97,78%. Clustering analysis with unweighted pair group with arithmetic average (UPGMA) method resulted dendrogram that exhibiting similarity values among individuals in range between 61% - 87% and clustered into two main clusters at 61% similarity value. The first cluster consists of samples taken from from Dlingo, Ngawen and Pracimantoro which has elevation above 200 m asl, while the second cluster consists of samples originating from Berbah which has elevation below 200 m asl. Amplification used 4 ISSR primer performed 2 monomorphic loci and 82 polymorphic loci. Monomorphic at 300 bp showed by ISSR2 and at 1800 bp showed by ISSR7 may be a candidate locus identifier species of A. atlas.Keywords: Attacus atlas, genetic variation, PCR-ISSR

Abstract

Thalassemia is an autosomal recessive genetic mutation disorder with symptoms similliar to anemia that causes deficiency synthesis of the globin chains (hemoglobin component inside erythrocytes). Thalassemia is classified based on secondary protein structure abnormalities in α globin protein or β globin protein. Based on data from Indonesian Thalassemia Foundation (YTI) in Indonesia, people with thalassemia, especialy β thalassemia is constantly increasing 8 to 10% per year, so it is very impotant to have a strategy that reduce the increasing number in the population. Genetic examination on the individual is very effective to detect quickly the person with thalassemia trait carrier, to reduce the number of thalassemia carrier population. Identification of 1st exon β globin gene mutations with sequencing method is one of the way to know the specific mutation in thalassemia carriers. Identification results can be used as a reference for the rapid detection of thalassemia trait carrier. This study aims to determine the type of mutation and location of nucleotide mutations in 1st exon β globin gene on carrier of thalassemia and the changes of amino acid translated by the mutant gene. The study was conducted by isolating the genome from the β thalassemia carrier blood, amplifying and sequencing the 1st exon β globin gene. The location of point mutation analysis and the changes of amino acid, was analyzed using computational method by comparative alignment using normal β globin gene as a comparison. The results of the research showed there is a point mutation in the 59th nucleotide caused by transition (T to C) and 147th nucleotide caused by transversion (G to C). The mutation type found on this study was a silent mutation because there is no change in the translated amino acid.Keywords: β globin, exon 1, mutation, thalassemia

Abstract

The tomato sauce is a complementary food that is often served with other foods, such as chicken noodles. Various factors can be the cause of contamination of tomato sauce by microbes like bacteria. These factors include environmental conditions that are not clean or container that was left open. Bacterial contamination in tomato sauce can result not suitable for consumption. Given by the high public interest in the consumption of tomato sauce with chicken noodle, it is necessary to observe the bacterial contamination of chicken noodle tomato sauce. To determine the bacterial contamination in tomato sauce of chicken noodle in traditional market of Condong Catur, Sleman, Yogyakarta. The samples used were tomato sauces that are used by chicken noodle stalls in the Condong Catur traditional market. Sampling was also carried out by recording the environmental conditions stalls and tomato sauce serving. To determine the bacterial contamination, samples examined in the laboratory based on the total bacterial count method using plate count agar (PCA) media. Seven of the ten samples showed the total number of bacteria <104 CFU/ml, whereas the other three samples are not suitable for consumption by the total number of bacteria >104 CFU / ml. The main factors of high contamination is poor hygiene of chicken noodle stall environmental, tomato sauce containers that are less clean and are often left open.Keywords: tomato sauce, total bacteria

Abstract

A research has been conducted on the antibacterial activity of Strychnos ligustrina seeds against Staphylococcus aureus ATCC 25923 in vitro. Strychnos ligustrina seeds extracted using two solvents, namely chloroform (chloroform extract) and 70% ethanol (ethanol extract) and analysis of the chemical content of the extract by thin layer chromatography and paper chromatography. The results of this study are expected to provide data activity of Strychnos ligustrina seeds as antibacterial scientifically. In this study, the chloroform extract and ethanol extract of Strychnos ligustrina seeds obtained by the filtration method with Soxhlet apparatus. Antibacterial activity test of chloroform extract and ethanol extract 70% from Strychnos ligustrina seeds performed using liquid dilution method, the parameters used are the MIC (minimum inhibitory concentration) and MKC (minimum killing concentration). MIC is determined by turbidity or clarity of the test solution, while MKC determined by scraping the respective concentrations of the extract / test solution on blood agar medium. The results showed that the chloroform extract and ethanol extract of Bidara Laut seed has antibacterial activity against Staphylococcus aureus ATCC 25923 and Salmonella thypi. MIC of each test extracts (chloroform and ethanol 70%) could not be determined because both of the test extract produce turbid color, while the MIC of the second test extract was 3.62% b/v for Salmonella thypi, 8.75% for Staphylococcus aureus ATCC 25923 (Strychnos ligustrina seeds chloroform extract), 22.5% for Salmonella thypi and 15% for Staphylococcus aureus ATCC 25923 (Strychnos ligustrina seeds ethanol extract). Results of thin layer chromatography and paper chromatography showed that the chloroform extract seeds contain alkaloids and steroids / triterpenoids, whereas the 70% ethanol extract contains alkaloids, tannins and steroids/triterpenoids. Keywords: antibacterial, chloroform extract, ethanol 70% extract, Strychnos ligustrina seeds, Salmonella thypi, Staphylococcus aureus

Abstract

Melon (Cucumis melo L.) is one of the fruits commodity priorities and require more attention. Plant breeding efforts led to improved varieties like TACAPA .TACAPA is a new melon cultivar derived from crosses between ♀ PI and ♂ Action with cross test PI. Since 2012, superior melon reassembled from crosses between ♀ TACAPA and ♂ PI become TANIA and ♀ TACAPA and ♂ Action become TALITA which projection can growth in critical lands karst. The objective of this research was to know ability to grow and morphological characteristics TALITA and TANIA melons were tested to determine nutrient levels of CaCO3. This research started from a field survey conducted in Hills of Mountain Sewu, South Mountain hills Sentolo Kulon Progo, and Dlingo Bantul, Yogyakarta.Melons grown on soil with fertilizer mixed media which further watering treated CaCO3 solution with a concentration of 0.5 % , 1%, 3%, 4%, and 5%. Observations were made until the time of harvest (± 10 weeks). TANIAand TALITA melon could grow on critical lands karst compared with parental. TANIA melonfruits have a weight, round, D - horizontal, D - vertical lower than two parental. TALITA melon fruits have a weight, D-horizontal higher than two parental, and D-vertical is lower than two parental.Keywords: critical land karst, Cucumis melo L., TALITA, TANIA

Abstract

This study aims to determine the hepatotoxic effects of extract ethanol Herba Comfrey the rabbit test animals by measuring levels of SGOT and SGPT. This study used 12 rabbits were divided into 4 groups. The control group (-) given Na.CMC 1% w/v, group I, II, and III were given suspensions of extract ethanol Herba Comfrey with each concentration of 2% w/v, 4% w/v and 8% w/v for 7 days. The results showed that the extract ethanol Herba Comfrey with concentration of 2% w/v, 4% w/v, and 8% w/v effects can cause hepatotoxicity. It is showed from the increase in the average SGOT value in rabbits at 13.96 U/L, 20.63 U/L and 37.13 U/L. and SGPT value of 20.73 U / L, 32.4 U / L and 44.66 U / L. extract Ethanol Herba Comfrey with concentration 8% w / v causes an increase in SGOT and SGPT concentrations higher than the other, and accelerate the liver damage or hepatotoxicity. Abstract

Passion fruit (Passifloraceae) was derived from tropical South America. Kind of passion fruit that widely cultivated in South Sulawesi is purple passion fruit (Passiflora edulis). This study aimed to observe the activity of Passiflora edulis chromosome at different stages of cell division and observed active mitosis time. The study was conducted by using exploratory descriptive study and obtained results a mitotic on Passiflora edulis occurs at 09.00 until 15.00 pm. Prophase seen in 5 minute after observation, prometaphase in 80 minute after the first observation, metaphase in 154 minutes after the first observation, anaphase in 227 minutes after the first observation, and telophase in 317 minutes after the first observation.Keywords: Chromosome, Mitosis, Passiflora edulis, Purple Passion Fruit

Abstract

Research on the effect of dose variations of manure on God’s Crown (Phaleria macrocarpa (Scheff.) Boerl.) growth was implemented in Ambarketawang, Gamping, Sleman, Yogyakarta. This study aimed to determine the effect of administration of dose variations of manure on God’s Crown growth. This research method using a complete randomized design (CRD) with one treatment factors which is administrating manure into polybags containing 2 kg of soil with dose variations of 0 g, 50 g, 100 g, 150 g, 200 g, and 250 g. The parameters measured in this study are height, leaf number, and fresh weight of Crown’s God plant. The results were analyzed by analysis of variance (ANOVA), ifthe data is meaningful (significant difference) then followed by LSD test level of 5%. The results obtained are the effects of administration of manure dose variation produce different Crown’s God (Phaleria macrocarpa (Scheff.) Boerl.) growth, and the best dose for Crown’s God (Phaleria macrocarpa (Scheff.) Boerl .) growth is 250 grams.Keywords: crown’s god (Phaleria macrocarpa (Scheff.) Boerl.), dose of manure

Abstract

Indole-3-acetic acid (IAA) is a growth hormone produced by plants. Several types of yeast has been reported to produce these hormones, and generally produce IAA in an higher amount than in plants. The purpose of this study is to get the yeast producing indole-3-acetic acid in the rhizosphere soil orchid Pecteilis susannae (L.) Rafin and to test the ability of yeast isolates to form IAA on medium L-tryptophan given as precursors. Yeasts were isolated and tested for their ability to produce IAA in vitro. The results showed that there are four potential yeast isolates that produce IAA which are isolates K2, U5, A3, and A1. Highest concentration of IAA was produced by A3 isolates grown on a medium containing GPB 200 ug / ml of L tryptophan in the amount of 56.262 g/ml.Keywords: Indole-3-acetic acid, rhizosfer yeast, Pecteilis susannae
